Bengkal plants can live in wetland ecosystems and have the ability in the phytoremediation process. Plants have an association with the diversity of microorganisms in the rhizosphere. Approximately 99% of all microorganisms in the environment can not be cultured so in analyzing the diversity of bacterial communities that can not be cultured conducted metagenomic analysis using the gene 16S rRNA. Metagenomic analysis is conducted using the latest linking technology, namely Next Generation Sequencing (NGS) which provides information that is considered effective and efficient such as finding annotations (naming), genome mapping, and further sequencing analysis. The rhizosphere region has different types of bacteria with largely unknown functional potential. Limited information about plant rhizosphere microbes that exist today, so conducted research on rhizosphere microbes to study the interaction between plants and soil microbes, understand various plant-related processes. This study aims to find out the genus of bacteria found in rhizospheres of bengkal plants based on the sequencing profile of the 16S rRNA gene. This research was conducted in January to July 2021 at the Laboratory of Genetics and Biotechnology Department of Biology Faculty of Mathematics and Natural Sciences Sriwijaya University. The stages of research conducted are soil sampling, genome DNA extraction, DNA quantity measurement, DNA extraction electrophoresis, gene 16S rRNA amplification, and high throughput sequencing data analysis. The conclusion of this study is a genus in rhizosphere bacteria bengkal plant (Nauclea orientalis L.) with gene sequence profile 16S rRNA identified as many as 15 genera with 14 genera including Achromobacter, Escherichia-Shigella, Enterobacter, Brevibacillus, Paenibacillus, Bacillus, Aneurinibacillus, Lysinibacillus, Clostridium, Acidothermus, Candidatus_Jorgensebacteria, Candidatus_Solibacter, Ochrobactrum, 1 genus of the order Saccharimonadales, and 1 genus are not classified.
