This study aims to determine the antioxidant activity of purple sweet potato leaf ethanol fraction against decreased levels of malondialdehyde (MDA) blood plasma and mouse pancreas in vivo by TBARS method. The ethanol fraction of purple sweet potato leaves is obtained using a multilevel maceration method using n-hexane solvent, ethyl acetate and ethanol 96%. The test animals were grouped into 6 groups, where the normal group was given only na-cmc, negative control was given aloksan 150 mg/kgBB, positive control was given vitamin C 9.9 mg/kgBB, while group I,II and III were given ethanol fractions of purple sweet potato leaves 50, 100 and 200 mg/kgBB respectively. The results of this study showed that the ethanol fraction of purple sweet potato leaves at doses of 50, 100 and 200 mg/kgBB has an effect as an antioxidant because it can lower the MDA levels of rat blood plasma by 2,028 ±0,025; 1,949±0.025 and 1,860±0.051 nmol/mL and pancreatic homogenate MDA levels of 2,042±0.052; 2,013±0.067 and 1,845±0.034 nmol/mL. Duncan's statistical analysis showed that there was no significant difference between doses of 50, 100 and 200 mg/kgBB with positive control group (vitamin C 9.9 mg/kgBB). so it can be said that the best dosage as an antioxidant is a dose of 50 mg/kgBB.
