RESPONS EMBRIOGENESIS MIKROSPORA PADI KULTIVAR SIAM PADA SUHU DINGIN DAN MEDIUM B DENGAN LAMA INKUBASI YANG BERBEDA SECARA IN VITRO
The research about "Embryogenesis Response of Rice Microspore Cultivar Siam of Cold Temperature and B Medium with Different Times of Incubation by In Vitro" was done in September 2010 to Juli 2011 in the Laboratory of Plant Physiology, Department of Biology, Faculty of Mathematics and Natural Sciences, University of Sriwijaya, Inderalaya. The aims of this research is to determine the response of microspore in cold temperature stress 5°C, B medium, the combination of cold temperatures 5°C and B medium by observation of viable and embryogenic microspore. This research used Group Randomized Design (GRD) with four treatments and four groups. The treatments were: control, cold temperature 5°C, B medium, the combination of cold temperatures 5°C and B medium. The groups were different times of incubation. They were 2, 4, 6, and 8 days. The results of this research showed that viable microspores at cold temperatures 5°C reached 63.25% and 2.5% to induced embryogenic microspores. Viable microspores in B medium treatment reached 66.36% and could to induced embryogenic microspores 26.5%. The treatment combination of cold temperatures 5°C and B medium, reached 89.75% microspores viable and most effective for inducing embryogenic microspores with an optimal incubation time for 4 days (81%). Characteristics of the morphology of embryogenic microspores, microspores are larger in size, ffagmented vacuole with the nucleus in the middle to form a starlike structure.
