UJI AKTIVITAS ANTIOKSIDAN FRAKSI ETIL ASETAT DAUN UBI JALAR UNGU (IPOMOEA BATATAS L.) PADA TIKUS PUTIH JANTAN GALUR WISTAR DIINDUKSI ALOKSAN
Free radicals can cause oxidative stress, due to an imbalance between oxidants and antioxidants that have the potential to cause cell damage. The results of phytochemical screening showed that the ethyl acetate fraction of purple sweet potato leaves contained secondary metabolites such as flavonoids and tannins which have antioxidant properties. The purpose of this study was to determine the characteristics and antioxidant activity of the ethyl acetate fraction of purple sweet potato leaves in vivo in reducing plasma malondialdehyde (MDA) levels and rat pancreatic homogenates. Purple sweet potato leaf fraction was obtained by graded maceration method using n-hexane and ethyl acetate as solvents. Rats were divided into six groups, namely normal control (NaCMC 1%), negative (alloxan 150 mg/kgBW), positive (vitamin E 120 IU/day), and the treatment group with doses of 75, 150, and 300 mg/kgBW. Antioxidant activity testing was carried out by measuring plasma MDA levels and pancreatic homogenates by UV-Vis spectrophotometry using the TBARS method. The data obtained were analyzed using the one way ANOVA test and followed by the Duncan test. The results showed that the doses of 75, 150, and 300 mg/kgBW body weight had antioxidant effects because they could reduce plasma MDA levels by 3,675 ± 1,003; 2,260 ± 1,476; and 2,160 ± 0,309 nmol/mL while for the homogenate levels of the pancreas decreased 4,145 ± 1,198; 2,905 ± 0,818; and 2,200 ± 0,309 nmol/mL. The ethyl acetate fraction of purple sweet potato leaves that has the best antioxidant effect and effect is a dose of 300 mg/kgBB because it has an antioxidant effect that is almost the same as vitamin E at a dose of 1,579 mg/200 gBW (2,075 ± 0,207 nmol/mL).
