Ethanolic extract of mangosteen peels has known to contain xanthone flavonoids which have anti-inflammatory effects and it will accelerate the process of fibroblast proliferation. The combination with aloe vera extract has expected to increase wound healing activity because it contains tannins and saponins. The delivery of the active substance in the two extracts will be more effective if it's formulated in a pharmaceutical preparation, one of them is a patch. The important component of the patch preparation is the polymer, so in this study a polymer in the form of chitosan combined with sodium alginate. The purpose of this study was to determine the effect of variations in sodium alginate concentration on the characteristics of the patch. Patches were made by ‘layer by layer’ method in three formulas namely F1, F2, and F3 with sodium alginate concentration to be used is 42.1 mg; 84.2 mg; 126.3 mg. The evaluations that to be done i.e organoleptic, weight uniformity, thickness uniformity, folding endurance, pH measurement, water content measurement, content uniformity, swelling test, FTIR, SEM, and in vitro permeation studies. The best formula found in F2’s patch which is indicated by a flexible, brownish-colored patch with a smooth surface. In addition, the patch also has a uniform weight, qualified of thickness 300 folds, has a pH of 4.3, and low water content of 3.99%. The swelling value in water, 95% ethanol and 99% ethanol, respectively was 4.3; 0.62; and 0.1, qualified for uniformity content and the release profile of the penetrated active substance of 38.94%. Morphological results from the patch preparation obtained an average pore diameter of 7,07 µm. Meanwhile, the results of the IR's spectra showed that the interaction of cross-linked chitosan, sodium alginate, and CaCl2 at a wave number of 1708,47 cm-1.
