Jackfruit leaf extract (Artocarpus heterophylus Lam.) was used as an active substance in acne treatment. Secondary metabolites contained flavonoids, saponins and tannins which have antibacterial activity. Flavonoids are thermolabile and easily oxidized so ethanol extract of jackfruit leaves is made in form of submicro particles. Micro particle jackfruit leaf extract with a size 5,485 microns were able to increase drug delivery to sebaceous glands where there colonies of Propionibacterium acnes. Preparation chosen was a gel, variations made on propylene glycol as a humectant in order to determine the effect of variations in propylene glycol concentration 10%, 12%, and 14% on physical tests, stability and diameter inhibition zone against Propionibacterium acnes. Antibacterial activity test using well method, by looking at the inhibition zone formed. Test results showed that preparation was able to inhibit Propionibacterium acnes with clear zone diameters of formula 1, formula 2, formula 3 respectively 8.66±1.1527 mm, 11.33±1.5270 mm, 15.33±4 .5092 mm. Increased diameter inhibition zone is directly proportional to the increase propylene glycol concentration. Physical tests including homogeneity, pH, viscosity, dispersibility, adhesion, washability, and stability tests results homogeneous; 5.16±0.02; 2867.66±180.15 cps; 5.78±0.02 cm; 10.82±0.17 seconds; 14.46±0.42 mL and stable. Increased propylene glycol concentration affects decrease in viscosity, washability and adhesion and increases spreadability. Results of statistical tests on antibacterial activity, physical tests and stability showed that there was a significant difference between treatment groups except for pH test. Results showed that the best formula was in the treatment group III (F3) with a propylene glycol concentration of 14%. Ekstrak daun nangka (Artocarpus heterophylus Lam.) dalam penelitian ini digunakan sebagai zat aktif dalam pengobatan jerawat. Metabolit sekunder yang terkandung berupa flavonoid, saponin dan tanin yang memiliki aktivitas antibakteri. Flavonoid bersifat termolabil dan mudah teroksidasi sehingga ekstrak daun nangka dibuat dalam bentuk mikro partikel. Mikro partikel ekstrak daun nangka dengan ukuran 5,485 mikron mampu meningkatkan penghantaran obat ke kelenjar sebasea dimana terdapat koloni Propionibacterium acnes. Sediaan yang dipilih berupa gel, dimana dilakukan variasi propilen glikol sebagai humektan yang bertujuan untuk mengetahui pengaruh variasi konsentrasi propilen glikol 10%, 12%, dan 14% terhadap uji fisik, stabilitas dan diameter zona hambat terhadap Propionibacterium acnes. Uji aktivitas antibakteri dilakukan dengan metode sumuran, dengan melihat zona hambat yang terbentuk. Hasil uji menunjukkan bahwa sediaan tersebut mampu menghambat bakteri Propionibacterium acnes dengan diameter zona bening formula 1, formula 2, formula 3 berturut-turut sebesar 8,66±1,1527 mm, 11,33±1,5270 mm, 15,33±4,5092 mm. Hal ini menunjukan peningkatan diameter zona hambat berbanding lurus dengan peningkatan konsentrasi propilen glikol. Uji fisik yang dilakukan meliputi uji homogenitas, pH, viskositas, daya sebar, daya lekat, daya tercuci, dan stabilitas dengan hasil berturut-turut homogen; 5,16±0,02; 2867,66±180,15 cps; 5,78±0,02 cm; 10,82±0,17 detik; 14,46±0,42 mL dan stabil. Konsentrasi propilen glikol yang meningkat mempengaruhi penurunan viskositas, daya tercuci dan daya lekat serta meningkatkan daya sebar sediaan. Hasil uji statistika aktvitas antibakteri, uji fisik dan stabilitas menunjukkan bahwa terdapat perbedaan yang signifikan antara kelompok perlakuan kecuali pada uji pH. Hasil penelitian menunjukan bahwa formula terbaik pada penelitian ini yaitu pada kelompok perlakuan III (F3) dengan konsentrasi propilen glikol sebesar 14%.
