ISOLASI SENYAWA METABOLIT SEKUNDER DARI JAMUR ENDOFITIK Trichoderma sp AKAR BROTOWALI (Tinospora Crispa L) DAN UJI AKTIVITAS ANTIBAKTERINYA
It was done the isolation of secondary metabolites compound frora endophytic fungus Trichoderma sp. Brotowali roots (Tinospora crispa L.). Isolation was begun with the cultivation of fungus in a 4 L PDB media (Potato Dextrose Broth) for eight weeks, then the liquid medium was extracted with ethyl acetate. Extracts that obtained then was separated and purificated by using chromatography techniques. The pure compounds were determined by UV, IR, NMR spectroscopy and antibacterial activity test with disc diffiision method toward Escherichia coli, Shigella dysenteriae, Staphylococcus aureus and Bacillus subtilis. Two compounds were obtained (I and II) with a melting point of each 88-89 °C and 155-156 °C. Based on the spectroscopic data analysis, the proposed compound I was (5-hydroxy-2H-2okso-pyran-4-yl) methyl acetate and compound II was 5-hydroxy-4 (hydroxymethyl)-2H-pyran-2-one. Compound I showed antibacterial activity at a concentration of 2000 ppm toward S. Aureus and B. Subtilis with clear zone diameter of 6.17 mm and 6.25 mm. Compound II showed antibacterial activity at a concentration of 250 ppm toward E. coli and B.Subtilis with the clear zone diameter 7.50 mm and 7.25 mm. Based on these data, it was concluded both isolated compounds were active as antibacterial at weak category.
