Background: Xanthine oxidase is an enzyme that catalyzes the formation of uric acid. Empirically, many people have used the root of the yellow wood as a natural antihyperuricemia medicine. Phytochemical tests were carried out on the ethanol extract of the leaves and roots of Kayu kuning, both of which contained almost the same secondary metabolites. The leaves of Kayu kuning (Arcangelisia flava (L.) Merr.) contain secondary metabolites of flavonoids and alkaloids that have the potential to inhibit the action of the xanthine oxidase enzyme.
Objective: To determine the inhibitory effect of yellow wood leaf (Arcangelisia flava (L.) Merr.) on the xanthine oxidase enzyme.
Methods: This research is an in vitro laboratory study. Yellow wood leaves were extracted by maceration using 96% ethanol as solvent. Allopurinol was used as a positive control. The ethanol extract of Kayu kuning leaves was carried out by phytochemical tests which included tests for the content of flavonoids, alkaloids, tannins, terpenoids, steroids, saponins, and quinones. Then, the sample was tested for the inhibition of the xanthine oxidase enzyme by measuring its absorbance using UV-vis spectrophotometry. Data were analyzed using linear regression analysis and IC50 values.
Results: The ethanol extract of Kayu kuning leaves contained alkaloids, flavonoids, triterpenoids, saponins, tannins, and quinones, but did not contain steroids. The ethanol extract of the leaves of Kayu kuning had an IC50 value of 174,619 ppm and allopurinol had an IC50 of 24,035 ppm.
Conclusion: The ethanol extract of Kayu kuning leaves has less potential to exert an inhibitory effect on the xanthine oxidase enzyme.
