PEMBUATAN ASAM-3-OKSOURSOLAT DAN ASAM-3-OKSO OLEANOLAT DARI TRITERPENOID BUAH TEMBESU SERTA AKTfVITAS SITOTOKSIK TERHADAP SEL LEUKEMIA P-388 SECARA IN-VITRO
Oxidation of the hydroxyl group (C-3 -OH) of ursolic acid [oleanolic acid], isolated from methanolic extract of tembesu fruit, had been done using Jones’ reagent (C1O3/H2SO4/H2O) in acetone with 40% yield. Spectrum data of ]H-NMR showed that the methine proton at C-3 position of the triterpenoids was no longer existed, and the analysis of mass spectrum showed that the oxidation products had molecular weight of 454 which was apropriate for compounds with moiecular formula C3oH46035 DBE = 8. These oxidation products, 3-oxoursolic acid [3-oxooIeanolic acid], with IC50 value of 18.5954 |ig/mL had cytotoxic activity against leukemia murine P-388 2.8 fold more active than the parent compounds, ursolic acid [oleanolic acid], with IC50 value of 53.4965 pg/mL.
