UJI EFEK ANTIKOLESTEROL EKSTRAK AIR DAUN KAYU KUNING (Arcangelisia flava) SECARA IN VITRO
Backgrounds. Cholesterol levels are the main factor in the formation of blood vessel plaque (artherosclerosis) which can cause an increase in blood pressure and blood vessels. Indonesian people have long used plant decoction as a means of maintaining health. This study aims to determine the anticholesterol effect of the aqueous extract of the leaves of Kayu kuning (Arcangelisia flava). Methods. This study is an in vitro experimental study with a posttest only control group design. The sample in this study was the aqueous extract of the leaves of Kayu kuning (Arcangelisia flava) using the infusion method with concentrations of 300 ppm, 600 ppm, and 1200 ppm. In this research, secondary metabolite compounds and anticholesterol effects were tested. Anticholesterol effect test was measured using the Liebermann-Burchard method with the aid of a spectrophotometric device to measure the percentage decrease in absorbance of standard cholesterol solutions and samples. Results. The results of the percentage reduction in cholesterol levels from the 3 concentrations of the extract solution were: 300 ppm (26.63%); 600 ppm (28.32%); 1200 ppm (54.24%). From the data on the percentage reduction in cholesterol levels, the IC50 value of the water extract of the leaves of kayu kuning (Arcangelisia flava) was 1.119.35 ppm. Analysis. The decrease in cholesterol levels from this study was caused by secondary metabolites contained in the water extract of Kayu kuning leaves in the form of flavonoids, tannins, and terpenoids which bind directly to cholesterol result in cholesterol levels measured by spectrophotometry are reduced. Conclusion. The water extract of the leaves of Kayu kuning (Arangelisia flava) has an anticholesterol effect in vitro and contains secondary metabolites such as alkaloids, flavonoids, saponins, tannins, and terpenoids.
