One of the biodiversity that has the potential to processed into medicinal ingredients are robusta coffee beans. Robusta coffee beans have antioxidant activity because it contains various polyphenol compounds, one of its polyphenols is chlorogenic acid. However, secondary metabolites that extracted from plants tend to be unstable in the preparation especially during storage. Therefore, robusta coffee bean extract preparation was carried out using PLGA polymer and PVA stabilizer. This research aims to improve the stability of secondary metabolites in the preparation. Submicro particle suspension was made contains of robusta coffee bean from ethanol extraction, PLGA and PVA, using variations in the concentration of PVA stabilizers of 50 mg, 100 mg, and 150 mg. The best submicro particel formulation of robusta coffee bean extract was obtained at concentration of PVA stabilizer of 50 mg, with percent encapsulation efficiency of 91.443%. The results of the stability test from submicro particle robusta coffee bean extract using the heating cooling cycle method with various buffer pH 2; 5.5; and 7.4 caused decrease of polyphenols content up to 33.752%, 23.371%, 16.848% respectively. The results of submicro particle characterization resulted in a particle diameter of 187.16 nm, poly dispersity index of 0.217 and zeta potential of -12.3 mV. Measurement of antioxidant activity using the DPPH (2,2-diphenyl-1-picrylhydrazyl) method resulted in the IC50 value of vitamin C compounds, robusta coffee bean ethanol extract, submicro particles of PLGA carrying robusta coffee ethanol extract before the stability test and after the stability test sequent values are 9.21 μg / mL; 61.91 μg / mL; 145.35 μg / mL; 209.02 μg / mL.
