Based on the results of initial observations, there were refill drinking water depots that did not meet the sanitation hygiene standards in the aspect of the place. Based on data from the Prabumulih City Health Office, in Prabumulih city there are 127 refill drinking water depots and the working area of the Prabumulih Timur Health Center there are 15 refill drinking water depots with 2,427 people consuming drinking water using refill drinking water depots. The purpose of this study was to analyze the hygiene and sanitation of refill drinking water depots in East Prabumulih District. This research is a qualitative research with descriptive method. Informants of this study amounted to 33 people. Collecting data using direct observation and in-depth interviews and for drinking water quality using the MPN method. The results of the study were based on direct observation of the hygiene and sanitation of drinking water depots, all depots met the requirements. Based on the quality of drinking water, there is 1 drinking water depot that does not meet the requirements. The conclusion is that the overall value of the variable at the refill drinking water depot is 71-85 and the quality of drinking water and there is 1 drinking water depot that contains Ecoli bacteria. Suggestions for checking regularly for refill drinking water quality so that the water produced does not contain Ecoli bacteria and is suitable for public consumption
