GAMBARAN POLIMORFISME TITIK -308 PROMOTER GEN TNF-Α DAN KADAR ANTIBODI PASCA PEMBERIAN VAKSIN INAKTIF COVID-19 DOSIS KEDUA
COVID-19 vaccination is a COVID-19 long-term prevention and control strategies which currently done by the government. COVID-19 vaccination is useful for inducing significantly neutralizing antibody levels and triggering a memory response to SARS-CoV-2. The effectiveness of vaccination is influenced by various factors, one of which is genetics. Previous studies have reported the association of -308 TNF-α gene promoter polymorphism with increased production of TNF-α as a central mediator of immune response. This study aimed to describe the -308 TNF-α gene promoter polymorphism and antibody concentration post second dose COVID-19 vaccination with inactivated vaccine. This research is an observational descriptive study using PCR-RFLP method and primary data collection of IgG s1 RBD SARS-CoV-2 concentration. The sample was Palembang citizen from several institutions who had been vaccinated with the second dose of inactivated COVID-19 vaccine on March – August 2021. Data analysis was carried out using SPSS 25 to assess the frequency distribution of genotype and allele at -308 TNF-α gene promoter and antibody concentration. The results showed a total of 55 samples were identified. Most subjects were 17-25 years old (43.6%) and female (52.7%). From the PCR-RFLP results, the genotype frequency distribution of GG 85.5%, GA 12.7%, and AA 1.8%. The G allele was found in 91.8% and the A allele was 8.2%. Meanwhile, data on antibody concentration using CMIA method showed 43.6% had high antibody concentration and 56.4% had low antibody concentration.
