Introduction: Plants that have been known to have an inhibitory effect on the alpha-glucosidase enzyme are the Benalu Kersen Leaves (Dendrophtoe pentandra (L). Miq) where this plant contains active compounds of flavonoids, alkaloids, terpenoids, steroids, tannins, and saponins. This study was conducted to validate the efficacy of the polar fraction of the ethyl acetate extract from benalu kersen leaves as a basic ingredient for drug development for diabetes by testing the inhibitory effect of the alpha-glucosidase enzyme in vitro. Methods: Simplisia extraction with ethyl acetate solvent followed by a fractionation process using a mixture of ethanol and ethyl acetate solvents with 5 different concentrations. The inhibitory effect of alpha-glucosidase enzyme was tested on all fractions and positive control acarbose. The absorbance value obtained was used to calculate the percentage inhibition of the alpha-glucosidase enzyme. The results of the percentage of inhibition will be carried out by linear regression analysis to get the IC50 value which is then categorized as very active, active or inactive. Result: The polar fraction of the ethyl acetate extract from benalu kersen leaves contains active compounds of alkaloids, flavonoids, steroids, saponins, and tannins. The IC50 value of the polar fraction of the ethyl acetate extract from benalu kersen leaves was successively according to a mixture of 95% ethanol solvents: ethyl acetate (9:1), (7:3), (5:5), (3:7), and (1: 9) are 61.10 ppm, 56.25 ppm, 45.15 ppm, 58,44 ppm, and 53.24 ppm which are all classified as active as inhibitors of the alpha-glucosidase enzyme. The positive control acarbose was classified as active as an inhibitor of the alpha-glucosidase enzyme with an IC50 value of 36.4 ppm. Keywords: Benalu kersen leaves, Dendrophtoe pentandra (L.)
