Insomnia often interferes with daily life in society, with an uncertain prevalence. One of the causes of insomnia is inflammation of the hypothalamus of the brain, which can damage various pathways of mechanisms and levels of brain neurotransmitters, which play a role in the sleepwake process. Benzodiazepines, a class of drugs for the treatment of insomnia, have a lot significant side effects. So, traditional herbs such as cinnamon extract (CE) can be used as an alternative because of their content which has the ability to work as anti-inflammatory and antioxidant for hypothalamic damage that occurs. This research is an in vivo experimental study with 30 white rats of the Wistar strain obtained from Eureka Research Laboratory Palembang as the research objects. The CE was made by simplicia maceration obtained from B2P2TOOT Tawangmangu, Central Java. The test animals used were 30 white rats of the Wistar strain. Rats were divided into 6 treatment groups which were divided into: normal control group (without PCPA induction), negative control (PCPA + Na-CMC), positive control (PCPA + estazolam 0.1 mg /kgBW), CE I (PCPA + 25 mg/kgBW), CE II (PCPA + 50 mg/kgBW), and CE III (PCPA + 100 mg/kgBW). PCPA was used to induce insomnia in rats for the first two days with the dose of 300 mg/kgBW by peritoneum injection. Estazolam and CE were given for the next 7 days as an intervention using a gastric sonde. The rats were then euthanized, brain tissue was removed, and their GABA levels were checked using ELISA. The results showed that the use of cinnamon extract in the CE I, CE II, and CE III groups was effective in increasing GABA levels in white rats of the Wistar strain of insomnia model. The ability of CE doses of 50 mg/kgBW and 100 mg/kgBW proved to be able to exceed the ability of estazolam at a dose of 0.1 mg/kgBW in increasing hypothalamic GABA levels. It can be concluded that Cinnamomum burmannii extract is effective in increasing hypothalamic GABA levels in Wistar strain white rats with insomnia model. Therefore, CE was effective in increasing hypothalamic GABA levels in insomniac Wistar rats.
