PRODUKSI ENZIM SELULASE DARI KAPANG Trichoderma reesei dan Aspergillus niger MENGGUNAKAN SUBSTRAT AMPAS TEBU UNTUK PEMBUATAN BIOETANOL
It has been conducted a research about bioethanol production from bagasse. Cellulose content in baggasse can be used as substrate in production of cellulase enzyme by using A. niger and T. reesei with solid State fermentation. It was carried out by variation of fermentation time at 27°C for 1, 3, 5 and 7 days. Concentration cellulase enzymes extract were analyzed by Lowry method. Whereas enzyme activities were analyzed by DNS method. This research was done in five steps; delignification, production of cellulase, hydrolysis, fermentation and distillation. The results of research showed that delignification with 4% NaOH concentration could reduce lignin as much as 25,21%. The highest protein content and enzyme activities were produced by A. niger as much as 0,304 mg/mL and 2,99 IU/mL for 5 days fermentation time and the highest protein content and enzyme activities were produced by T. reesei as much as 0,278 mg/mL and 2,31 IU/mL for 3 days fermentation time. The highest glucose concentration content of hydrolysis for 3 days incubation time which were produced by T. reesei as much as 8,367 mg/mL. The highest ethanol content (distillation boiling point 78°C) of fermentation with anaerob facultative were produced by hydrolysis with cellulase enzyme from T. reesei as much as 17,613 % for 3 days fermentation time.
