OPTIMASI NANOEMULSI VIRGIN COCONUT OIL (VCO) DENGAN METODE DESAIN FAKTORIAL DAN UJI AKTIVITAS ANTIBAKTERI TERHADAP BAKTERI STAPHYLOCOCCUS AUREUS SECARA IN VITRO
Virgin Coconut Oil (VCO) contains lauric acid which has the potential as an antibacterial, used against Staphylococcus aureus. The preparation of VCO nanoemulsion was carried out to increase its antibacterial effect. This research was conducted to optimize the VCO nanoemulsion preparation and to determine its antibacterial activity against Staphylococcus aureus bacteria compared to VCO without a nanoemulsion. Optimization was carried out using the 22 factorial design method through Design Expert 13® on the response of pH, specific gravity, percent transmittance, particle size, and particle distribution. The optimum formula of the VCO nanoemulsion that has been obtained was evaluated by centrifugation test, cycling test, determination of the diameter of the inhibition zone by disc diffusion method, minimum inhibitory concentration (MIC), and minimum kill concentration (MBC). Optimum formulation of nanoemulsion preparation VCO at a emulsifier concentration using Tween 80 40% and PEG 400 26%. The centrifugation test showed no phase separation.
